One persistent puzzle in the life sciences is the asymmetric lipid composition of the cellular plasma membrane: while the exoplasmic leaflet is enriched in lipids carrying predominantly saturated fatty acids, the cytoplasmic leaflet hosts preferentially lipids with (poly‐)unsaturated fatty acids. Given the high energy requirements necessary for cells to maintain this asymmetry, the question naturally arises regarding its inherent benefits. In this paper, we propose asymmetry to represent a potential solution for harmonizing two conflicting requirements for the plasma membrane: first, (...) the need to build a barrier for the uncontrolled influx or efflux of substances; and second, the need to form a fluid and dynamic two‐dimensional substrate for signaling processes. We hence view here the plasma membrane as a composite material, where the exoplasmic leaflet is mainly responsible for the functional integrity of the barrier and the cytoplasmic leaflet for fluidity. We reinforce the validity of the proposed mechanism by presenting quantitative data from the literature, along with multiple examples that bolster our model. (shrink)

In our daily lives we conceive of our surroundings as an objectively given reality. The world is perceived through our senses, and ~hese provide us, so we believe, with a faithful image of the world. But occ~ipnally we are forced to realize that our senses deceive us, e. g., by illusions. For a while it was believed that the sensation of color is directly r~lated to the frequency of light waves, until E. Land (the inventor of the polaroid camera) showed (...) in detailed experiments that our perception of, say, a colored spot depends on the colors of its surrounding. On the other hand, we may experience hallucinations or dreams as real. Quite evidently, the relationship between the "world" and our "brain" is intricate. Another strange problem is the way in which we perceive time or the "Now". Psychophysical experiments tell us that the psychological "Now" is an extended period of time in the sense of physics. The situation was made still more puzzling when, in the nineteen-twenties, Heisenberg and others realized that, by observing processes in the microscopic world of electrons and other elementary particles, we strongly interfere with that world. The outcome of experiments - at least in general - can only be predicted statistically. What is the nature ofthis strange relationship between "object" and "observer"? This is another crucial problem of the inside-outside or endo-exo dichotomy. (shrink)

Background:Clinical investigation is a growing field employing increasing numbers of nurses. This has created a new specialty practice defined by aspects unique to nursing in a clinical research context: the objectives, setting, and nature of the nurse–participant relationship. The clinical research nurse role may give rise to feelings of ethical conflict between aspects of protocol implementation and the duty of patient advocacy, a primary nursing responsibility. Little is known about whether research nurses experience unique ethical challenges distinct from those experienced (...) by nurses in traditional patient-care settings.Research objectives:The purpose of the study was to describe the nature of ethical challenges experienced by clinical research nurses within the context of their practice.Research design:The study utilized a qualitative descriptive design with individual interviews.Participants and research context:Participating nurses self-identified as having experienced ethical challenges during screening. The majority were Caucasian, female, and worked in outpatient settings. Approximately 50% had > 10 years of research experience.Ethical considerations:The human subjects review board approved the study. Written informed consent was obtained.Findings:Predominant themes were revealed: the inability to provide a probable good, or/do no harm, and dual obligations. The following patterns and subthemes emerged: conflicted allegiances between protocol implementation, needs of the participant, desire to advance science, and tension between the nurse–patient therapeutic relationship versus the research relationship.Discussion:Participants described ethical challenges specific to the research role. The issues are central to the nurse–participant relationship, patient advocacy, the nurse’s role in implementing protocols, and/or advancing science.Conclusion:Ethical challenges related to the specialized role of clinical research nurses were identified. More research is warranted to fully understand their nature and frequency and to identify support systems for resolution. (shrink)

The fundamental mechanisms of protein and lipid organization at the plasma membrane have continued to engage researchers for decades. Among proposed models, one idea has been particularly successful which assumes that sterol‐dependent nanoscopic phases of different lipid chain order compartmentalize proteins, thereby modulating protein functionality. This model of membrane rafts has sustainably sparked the fields of membrane biophysics and biology, and shifted membrane lipids into the spotlight of research; by now, rafts have become an integral part of our terminology to (...) describe a variety of cell biological processes. But is the evidence clear enough to continue supporting a theoretical concept which has resisted direct proof by observation for nearly twenty years? In this essay, we revisit findings that gave rise to and substantiated the raft hypothesis, discuss its impact on recent studies, and present alternative mechanisms to account for plasma membrane heterogeneity. Die Entscheidung für oder gegen lebensverlängernde Behandlungsmaßnahmen geht inzwischen der Hälfte aller Todesfälle in Europa voraus. Sie wird im klinischen Alltag häufig als ethische Herausforderung wahrgenommen, zudem sind unter Klinikern juristische Unsicherheiten und Fragen der korrekten Vorgehensweise verbreitet. Die hier vorgestellte Münchner Leitlinie zu Entscheidungen am Lebensende soll rechtliche Unsicherheit reduzieren, Klinikumsmitarbeiter für die ethische Dimension von Therapieentscheidungen am Lebensende sensibilisieren und ethisch begründete Entscheidungen fördern. Aus organisationsethischer Perspektive soll mit der Leitlinie eine Reflexion und Meinungsbildung zu einem ethisch relevanten (...) Themenbereich erfolgen und ein Beitrag zur Qualitätssicherung der Patientenversorgung und damit auch zur Patientenzufriedenheit geleistet werden. Die Prozesse der Leitlinienentwicklung, -aktualisierung und -implementierung werden vorgestellt. Die wesentlichen inhaltlichen Elemente der Leitlinie mit Darstellung der Rechtslage, der Definition zentraler Begriffe und Klärung medizinischer Entscheidungskriterien werden an Hand eines Entscheidungsalgorithmus dargestellt. Inhalt und Prozess der Leitlinienentwicklung werden auf die folgenden organisationsethischen Qualitätskriterien hin überprüft: deliberativer Prozess, inhaltliche Transparenz, repräsentative Zusammensetzung der Arbeitsgruppe, Implementierung und Evaluation der Leitlinie. Institutionen, die diese Leitlinie übernehmen wollen, sollten diese von einem autorisierten Gremium überprüfen lassen und auf die spezifischen Bedürfnisse ihrer Institution adaptieren. Our report and bibliography concentrate on research in the philosophy of science carried out in Austria within the last 20 years. The term 'philosophy of science' is here to be understood in the broad sense of 'Wissenschaftstheorie', that is, syntactics, semantics and pragmatics of the natural sciences and of the humanities, including law. After a general introduction to the philosophy of science scene in Austria, we report about those institutions in Austria at which relevant research has been conducted, starting with (...) institutions in Graz and then continuing - in alphabetical order - with institutions in Innsbruck, Klagenfurt, Linz, Salzburg, and Wien. Our report is supplemented by a bibliography; please note that this contains only references to original publications which deal mainly with questions in the philosophy of science, hence no contributions to lexica, no reviews, no translations, no articles in mass media, no editorial and no unpublished works are cited. Finally, there is an appendix, Alphabetical List of Austrian Institutions at which Philosophy of Science is Conducted, to facilitate communication between you and Austrian philosophers in whose work you may become interested by reading this report. ENGLISH ABSTRACT: Basic statements play a central role in Popper's "The Logic of Scientific Discovery", since they permit a distinction between empirical and non-empirical theories. A theory is empirical iff it consists of falsifiable statements, and statements (of any kind) are falsifiable iff they are inconsistent with at least one basic statement. Popper obviously presupposes that basic statements are themselves empirical and hence falsifiable; at any rate, he claims several times that they are falsifiable. In this paper we prove that (...) no basic statement is falsifiable, if the term 'basic statement' is to be understood according to Popper's definitions of that term. More precisely, we prove not only that no Popperian basic statement in the narrow sense of the word is falsifiable, but also that no Popperian basic statement in the broader sense, and even in the broadest possible sense, is falsifiable. This leads to the paradoxical result that, according to Popper's definitions of 'basic statement', the basis of the empirical sciences is not itself empirical. Furthermore, we point out a similar paradox as regards Popper's falsifiability scheme for theories. We close with a look at possibilities for overcoming these paradoxes. DEUTSCHE ZUSAMMENFASSUNG: Basissätze spielen eine zentrale Rolle in Poppers Logik der Forschung, denn sie erlauben die Unterscheidung zwischen empirischen und nichtempirischen Theorien: Eine Theorie ist empirisch genau dann, wenn sie aus falsifizierbaren Aussagesätzen besteht, und Aussagesätze (beliebiger Art) sind falsifizierbar genau dann, wenn sie mindestens einem Basissatz widersprechen. Popper setzt offensichtlich voraus, dassŸ die Basissätze selbst empirisch und somit falsifizierbar sind. Jedenfalls behauptet er mehrmals ihre Falsifizierbarkeit. Wir beweisen in unserem Aufsatz, dassŸ die Basissätze nicht falsifizierbar sind, und wir beweisen dies nicht nur für Poppersche Basissätze im engeren Sinn, sondern auch für Poppersche Basissätze im weiteren Sinn und schlieߟlich für Poppersche Basissätze im weitesten, mit den Vorstellungen Poppers gerade noch verträglichen Sinn. Dies führt zu dem paradoxen Ergebnis, dass nach Poppers eigenen methodologischen Postulaten die Basis der empirischen Wissenschaften nicht selbst empirisch ist. Darüber hinaus entwickeln wir ein ähnliches Paradoxon bezüglich Poppers Falsifizierbarkeitsschema für Theorien. Zum AbschlussŸ unseres Aufsatzes betrachten wir einige Möglichkeiten, die von uns entdeckten Paradoxa zu überwinden. (shrink)

This book describes in detail the relationship between symbolic representations of knowledge and abstract states of knowledge, exploring along the way the foundations of knowledge, knowledge bases, knowledge-based systems, and knowledge representation and reasoning. The idea of knowledge bases lies at the heart of symbolic, or "traditional," artificial intelligence. A knowledge-based system decides how to act by running formal reasoning procedures over a body of explicitly represented knowledge—a knowledge base. The system is not programmed for specific tasks; rather, it is (...) told what it needs to know and expected to infer the rest. This book is about the logic of such knowledge bases. It describes in detail the relationship between symbolic representations of knowledge and abstract states of knowledge, exploring along the way the foundations of knowledge, knowledge bases, knowledge-based systems, and knowledge representation and reasoning. Assuming some familiarity with first-order predicate logic, the book offers a new mathematical model of knowledge that is general and expressive yet more workable in practice than previous models. The book presents a style of semantic argument and formal analysis that would be cumbersome or completely impractical with other approaches. It also shows how to treat a knowledge base as an abstract data type, completely specified in an abstract way by the knowledge-level operations defined over it. (shrink)

A comprehensive, self-contained survey of the theory and applications of differential games, one of the most commonly used tools for modelling and analysing economics and management problems which are characterised by both multiperiod and strategic decision making. Although no prior knowledge of game theory is required, a basic knowledge of linear algebra, ordinary differential equations, mathematical programming and probability theory is necessary. Part One presents the theory of differential games, starting with the basic concepts of game theory and going on (...) to cover control theoretic models, Markovian equilibria with simultaneous play, differential games with hierarchical play, trigger strategy equilibria, differential games with special structures, and stochastic differential games. There is a plurality of formal constraints for aggregating probabilities of a group of individuals. Different constraints characterise different families of aggregation rules. In this paper, we focus on the families of linear and geometric opinion pooling rules which consist in linear, respectively, geometric weighted averaging of the individuals’ probabilities. For these families, it is debated which weights exactly are to be chosen. By applying the results of the theory of meta-induction, we want to provide a general rationale, namely, optimality, (...) for choosing the weights in a success-based way by scoring rules. A major argument put forward against weighting by scoring is that these weights heavily depend on the chosen scoring rule. However, as we will show, the main condition for the optimality of meta-inductive weights is so general that it holds under most standard scoring rules, more precisely under all scoring rules that are based on a convex loss function. Therefore, whereas the exact choice of a scoring rule for weighted probability aggregation might depend on the respective purpose of such an aggregation, the epistemic rationale behind such a choice is generally valid. Social scientific reasoning (SSR) is essential to social science education and to a democratic society as a whole. Students are challenged to analyze and reason about social problems such as social inequality, crime, and poverty. However, students experience difficulties with SSR. This study addresses the research question: Which design principles can guide teachers in designing lessons that promote social scientific reasoning? In this design-based research, four social science teachers employed a conceptualization of SSR and its levels together with three initial (...) design principles to develop curriculum materials and activities. These design principles and curriculum materials were piloted in two secondary education classes (9th and 10th grades) and evaluated by four social science teachers, four social science teacher educators and 90 students. The study produced six design principles that can promote students’ SSR. In combination with the curriculum materials, those design principles can help develop teachers’ pedagogical content knowledge and guide the design of tasks and units that develop SSR. (shrink)
